DETEKSI PEPPER VEIN YELLOWS VIRUS YANG BERASOSIASI DENGAN PENYAKIT YELLOW VEIN BANDING PADA TANAMAN MENTIMUN DI TABANAN - BALI

Abstract

Gejala menguning antara lamina daun (yellow vein banding) telah ditemukan pada tanaman mentimun di Bali sejak tahun 2014. Gejala pada tanaman mentimun tersebut memiliki kemiripan dengan gejala pada tanaman cabai yang disebabkan oleh infeksi Pepper vein yellow virus, anggota genus Polerovirus. Tujuan penelitian ialah membuktikan penyebab penyakit menguning antara lamina daun pada tanaman mentimun di Bali. Metode identifikasi menggunakan RT-PCR dan analisis sikuensing. Sepasang primer spesifik PeVYV-CP-F-BamH1/PeVYV-CP-R-Pst1 berhasil mengamplifikasi bagian protein selubung PeVYV dengan ukuran ± 650 pb. Analisis sikuen nukleotida menunjukkan bahwa isolat mentimun Bali memiliki kemiripan > 95% dengan isolat PeVYV asal Bali, dan Rembang (Indonesia), Jepang serta Yunani yang menginfeksi tanaman cabai.Kata kunci: Polerovirus, protein selubung, RT-PCR, sikuen nukleotida.
